Study on the Expression Changes and Effects of the Key Protein of the PGC-1α Pathway in the Intervention of Sini Decoction in Shaoyin Disease (Cardiac and Kidney Yang Deficiency)

Abstract

Objective: In this study, the effects of peroxlsome proliferator-activated re-ceptor- γ coactlvator-1α(PGC-1α)-nuclear respiratory factor-1(NRF-1)- mito-chondrial transcription factor A(TFAM) pathway signaling and the effect of Sini Decoction on PGC-1α-NRF-1-TFAM pathways and the effects of Sini Decoction on the PGC-1α-NRF-1-TFAM γ pathway were investigated by de-tecting the changes in the key proteins of the energy metabolism pathway of myocardial tissue in rats with cardiac-kidney-yang deficiency model and its corresponding period. Methods: SD rats are randomly divided into normal group at "Yujie time", normal group at "Non-Yujie Time", model group at "Yujie Time", model group at "Non-Yujie Time", drug group at "Yujie Time", and drug group at "Non-Yujie Time". After the end of molding and adminis-tration, the myocardial tissue was taken from “Yujie Time” and "Non-Yujie Time" for 6 days. Western Blot detected the levels of PGC-1α, NRF-1, TFAM protein in experimental rat myocardial tissue during the Yujie Time and Non-Yujie Time. Results: The gray value of PGC-1α protein in the model group within 1 to 6 days was basically lower than that of the normal group, and the gray value of NRF-1 and TFAM protein was higher than that of the normal group in the early stage and lower than that in the normal group in the later stage. The gray value of PGC-1α and NRF-1 proteins in the drug group was higher than that of the model group, and the gray value of TFAM protein was lower than that of the model group in the early stage and higher than that in the model group in the later stage. Conclusion: In the doxorubicin and hy-drocortisone-induced "heart-kidney yang deficiency" rat model, PGC-1α protein expression decreased, and NRF-1 and TFAM protein expression in the pre-course stage of the disease group were higher than in the normal group, suggesting that the high expression of NRF-1 and TFAM protein in the early stage of CHF disease may exert its stress protection mechanism, and the expression decreased in the later stage of the experiment, and we believe that the PGC-1α-NRF-1-TFAM signaling pathway may be conducted in the later stage of the disease course; Sini Decoction can upregulate the expression of PGC-1α and NRF-1 proteins in myocardial tissues, upregulate the late expression of TFAM proteins, or regulate the PGC-1α-NRF-1-TFAM signaling pathway.
